The base of the shower will have a shower pan to collect any water that gets through the caulk but you still don t want water getting through there and growing mold in the shower.

Due to this i recommend caulking the vertical wall joints first and then caulk the tub rim when you finish.
That way hopefully any water that may find its way down the wall runs behind the tile and discharges out through the wells.
